SARS-CoV-2 variant has Tyrosine (Y) amino acid at 501th location in Spike protein define as Y501. This amino acid change in the spike protein causes the strong binding of SARS-CoV-2 to the ACE2.

This mutation enhances ACE-2 receptor binding by the virus and interferes with the action of existing antibodies on themutated S-protein.

Diagnovital® SARS-CoV-2 K417N Mutation Detection Kit is a real-time RT-PCR-based test for the detection and discrimination of SARS-CoV-2 K417N mutation in respiratory specimens. This kit is designed to run only samples that have previously proven to be SARS-CoV-2 positive.

DIAGNOVITAL® SARS-CoV-2 K417N Mutation Detection Kit detects K417N mutation in extracted SARS-CoV-2 RNA from nasopharyngeal and oropharyngeal swab samples during infection. Positive results indicate the presence of SARS-CoV-2 RNA detects one single point mutation which result in the exchange of the amino acid Lysine to Asparagine in the spike protein at position 417. Mutations at position 417 is found in two variant lineages, however the exchange to an Asparagine is specific for South Africa lineage B.1.351 and can help the precise distinction of the different variant types of SARS-CoV-2. The K417N is a mutation which occurs in different lineages and has already been found in the South African (B.1.351) and Brazilian (B.1.1.28) lineage. The accompanying K417N and E484K mutations in the South African strain provide a counterpoise to the spike's increased affinity due to the N501Y mutation. They prevent the formation of two salt bridges that help to form and stabilize the RBD-ACE2 complex. This reduces ACE2 binding affinity. Thus, this strain is less infectious and less rapidly spreading than the UK strain. This is even though both share the latter substitution.The mutation is in the spike protein and appears to have an impact on the body’s immune response and, possibly, vaccine efficacy. Monoclonal and serum-derived antibodies are reported to be from 10 to 60 times less effective in neutralizing virus bearing the K417N mutation.

DIAGNOVITAL® SARS-CoV-2 L452R Mutation Detection Kit detects the L452R mutation in extracted SARS-CoV-2 RNA from nasopharyngeal and oropharyngeal swab samples during infection. Positive results indicate the presence of SARS-CoV-2 L452R mutation. The L452R mutation is characterized by a Leucine (L) to Arginine (R) shift at amino acid position 452. This mutation enhances ACE-2 receptor binding by the virus and interferes with the action of existing antibodies on the mutated S-protein. Additionally, L452R mutation is postulated to provide resistance to T-cells. L452R mutation contributes to escaping from HLA-A24 mediated cellular immunity and increasing binding affinity to ACE2 and viral infectivity. The mutant SARS-CoV-2 L452R has Leucine-452 is positioned in the receptor-binding motif of RBD, in the interface of direct contact with the ACE2 receptor. Its replacement with Arginine is predicted to result in both a much stronger binding to the receptor and escape from neutralizing antibodies.
